Original Contribution INCREASED LIPOPROTEIN OXIDATION IN ALZHEIMER’S DISEASE

چکیده
Oxidation has been proposed to be an important factor in the pathogenesis of Alzheimer’s disease (AD) and amyloid is considered to induce oxidation. In biological fluids, including cerebrospinal fluid (CSF), amyloid is found complexed to lipoproteins. On the basis of these observations, we investigated the potential role of lipoprotein oxidation in the pathology of AD. Lipoprotein oxidizability was measured in vitro in CSF and plasma from 29 AD patients and found to be significantly increased in comparison to 29 nondemented controls. The levels of the hydrophilic antioxidant ascorbate were significantly lower in CSF and plasma from AD patients. In plasma, -carotene was significantly lower in AD patients compared to controls while -tocopherol levels were indistinguishable between patients and controls. In CSF, a nonsignificant trend to lower -tocopherol levels among AD patients was found. Polyunsaturated fatty acids, the lipid substrate for oxidation, were significantly lower in the CSF of AD patients. Our findings suggest that (i) lipoprotein oxidation may be important in the development of AD and (ii) the in vitro measurement of lipid peroxidation in CSF might become a useful additional marker for diagnosis of AD. © 2000 Elsevier Science Inc. Glucose Influence on Copper Ion-Dependent Oxidation of Low Density Lipoprotein
Background: It is well established that oxidative modification of low density lipoprotein (LDL) plays a causal role in human atherogenesis and the risk of atherosclerosis is increased in patients with diabetes mellitus. We examined the in vitro effect of glucose on native and glycated LDL oxidation using copper ion dependent oxidation system. Methods: In this study, LDL was isolated from plasma...
متن کاملHypothesis Paper AMYLOID- : AN ANTIOXIDANT THAT BECOMES A PRO-OXIDANT AND CRITICALLY CONTRIBUTES TO ALZHEIMER’S DISEASE
Elevated production of amyloid(A ) as a preventive antioxidant for brain lipoproteins under the action of increased oxidative stress in aging is postulated to represent a major event in the development of Alzheimer’s disease (AD). Increase in A production is followed by chelation of transition metal ions by A , accumulation of A -metal lipoprotein aggregates, production of reactive oxygen speci...
متن کاملSerum Lipids and Hippocampal Volume: The Link to Alzheimer’s Disease?
The association between hippocampal volume (as a presumed index of Alzheimer’s disease pathology) with serum total cholesterol, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol was studied in 86 elderly subjects with a range of cognitive functions. High-density lipoprotein cholesterol, but not low-density lipoprotein cholesterol or total cholesterol, was associated ...
متن کاملAntioxidant Effects of Vitamins C and E on the Low-Density Lipoprotein Oxidation Mediated by Myeloperoxidase
Background: Oxidative modification of low-density lipoprotein (LDL) appears to be an early step in the pathogenesis of atherosclerosis. Meanwhile, myeloperoxidase (MPO)-catalyzed reaction is one of the potent pathway for LDL oxidation in vivo. The aim of this study was to evaluate in vitro antioxidant effects of vitamins C and E on LDL oxidation mediated by MPO. Methods: MPO was isolated from f...
متن کاملRelationship between High-Density Lipoprotein and C-reactive Protein in Patients with Atherosclerosis
Abstract Background and Objective: Atherosclerosis is an inflammatory disease mostly caused by oxidation of low density lipoproteins (LDLs) while High-density lipoproteins (HDLs) oppose atherosclerosis by inhibiting the oxidation of LDLs. Serum concentration of C- reactive protein (CRP) also increases as an acute phase protein in inflammatory conditions like atherosclerosis. We aimed to evalu...
